Ondansetron Hydrochloride Suppositories USP 4mg

Composition:

Each suppository contains:

  • Ondansetron Hydrochloride (equivalent to Ondansetron 4mg)

Description:

Ondansetron Hydrochloride is a selective 5-HT3 receptor antagonist used to prevent and treat nausea and vomiting caused by chemotherapy, radiation therapy, and surgery. The suppository formulation provides an alternative non-oral route of administration for patients unable to take oral medication.

Indications:

Ondansetron Hydrochloride Suppositories are indicated for the prevention and treatment of:

  • Chemotherapy-induced nausea and vomiting (CINV)
  • Radiation therapy-induced nausea and vomiting (RINV)
  • Postoperative nausea and vomiting (PONV)

Dosage & Administration:

Adults & Children (>12 years):

  • One suppository (4mg) inserted rectally every 8-12 hours as needed.

Children (4-11 years):

  • One suppository (4mg) inserted rectally every 12 hours.

Administration:

  • For rectal use only.
  • Wash hands before and after use.
  • Insert gently into the rectum.
  • Avoid excessive movement for a few minutes after insertion.

Contraindications:

  • Hypersensitivity to Ondansetron or other 5-HT3 antagonists.
  • Patients with severe hepatic impairment (dose adjustment required).

Warnings & Precautions:

  • Serotonin syndrome risk when used with serotonergic drugs (e.g., SSRIs, SNRIs).
  • QT prolongation risk, especially in patients with heart conditions.
  • Use with caution in patients with liver disease.
  • May cause constipation in some patients.

Adverse Reactions:

  • Common: Headache, dizziness, fatigue, constipation
  • Serious: Arrhythmias, serotonin syndrome, severe allergic reactions

Drug Interactions:

  • Apomorphine: Severe hypotension risk
  • Serotonergic drugs (SSRIs, SNRIs): Increased risk of serotonin syndrome
  • CYP3A4 inducers (e.g., rifampin): May reduce Ondansetron efficacy

Storage Conditions:

  • Store below 25°C (77°F)
  • Protect from heat and moisture
