Description:
Famotidine is a histamine H2-receptor antagonist that inhibits gastric acid secretion. It is used for the treatment of gastric and duodenal ulcers, gastroesophageal reflux disease (GERD), and Zollinger-Ellison syndrome.
Indications:
Famotidine for Injection is indicated for:
- Active duodenal ulcer
- Gastric ulcer
- Gastroesophageal reflux disease (GERD)
- Pathological hypersecretory conditions (e.g., Zollinger-Ellison syndrome)
- Prevention of stress ulcers in critically ill patients
Dosage & Administration:
Intravenous (IV) Use Only
- Administer as slow IV injection over 2 minutes or IV infusion over 15-30 minutes.
Adult Dosage:
- Peptic ulcer disease (PUD) & GERD: 20mg IV every 12 hours
- Zollinger-Ellison syndrome: Initial 20mg IV every 6 hours, titrated as needed
- Stress ulcer prophylaxis: 20mg IV every 12 hours
Pediatric Dosage (≥1 year):
- GERD & Peptic Ulcer Disease: 0.5mg/kg IV every 12 hours, max 40mg/day
Preparation & Administration:
- Slow IV push: Dilute in normal saline (0.9% NaCl) or 5% dextrose to final concentration 4mg/mL
- IV infusion: Dilute in 50-100mL of compatible IV fluid and infuse over 15-30 minutes
- Avoid rapid IV injection (risk of hypotension)
Contraindications:
- Hypersensitivity to famotidine or other H2-receptor antagonists
Precautions:
- Adjust dose in renal impairment (CrCl <50 mL/min)
- Use with caution in elderly patients (risk of CNS effects)
- Not recommended in pregnancy unless necessary
Adverse Reactions:
- Common: Headache, dizziness, constipation, diarrhea
- Serious: Bradycardia, hypotension, confusion (especially in elderly or renal impairment)
Drug Interactions:
- Reduced absorption of drugs requiring acidic pH (e.g., ketoconazole, atazanavir)
- May enhance effects of warfarin (monitor INR closely)
Overdose & Management:
- Symptoms: Hypotension, confusion, arrhythmias
- Treatment: Supportive care, symptomatic management
Storage Conditions:
- Store below 25°C, protect from light
- Use reconstituted solution within 24 hours (if refrigerated at 2-8°C)
